About usResearch in the Diabetes Laboratory of the Institute of Experimental Endocrinology (an EU

Centre of Excellence), Slovak Academy of Sciences has been focused over the years to elucidations of molecular mechanisms of the insulin resistance syndrome (IRS). Special attention has been paid to mechanisms of the favourable effect of dietary supplements and novel molecules with hypolipidemic effect on the individual components of the IRS. Recent clinical studies are focusing at the endocrine/paracrine function of the adipose tissue in relation to molecular mechanisms of insulin resistance development in obesity, as well as in association with growth hormone deficiency or chronic obstructive pulmonary disease. Various types of fat depots are investigated using different approaches of genomics and proteomics. In addition we are very interested in clinical situations associated with lack of adipose tissue or on the other hand with morbid obesity, which represent two completely distinct etiologies of insulin resistance development. Clinical research is going to be paralleled by animal studies focused at the nutritional, environmental and genetic determinants of obesity and diabetes using animal models with different susceptibility to dietinduced insulin resistance and obesity. The above mentioned studies are fulfilled in close colaboration with Laboratory of Human Endocrinology at the IEE, where the members of the Laboratory are physicians responsible for clinical phenotyping related to studied subjects. Their studies are focusing at neuroendocrine mechanisms in of insulin resistance, hypertension and immune processes in human as well as hormonal regulation of lipid and glucose metabolism in obesity, growth hormone deficiency and corticosteroid abundance.The available technologies include a) a whole variety of clinical physiology techniques allowing to assess the whole body in vivo insulin action and related tissue parameters in collected bioptic specimens; b) a broad range of standardised analytical techniques for measurement of hormone and antibodies plasma levels using automated electrochemiluminiscence and Elisa systems and c) molecular biology methods including RNA expression studies using the Real Time PCR, the mutations/polymorphisms as detected by RFLP or by bi-directional DNA sequencing using the capillary ABI PRISM genetic analyzer, as well as pre-screening approach by dHPLC

system.
ContributionsThe Gasperikova lab has access to patients with type 2 diabetes and obesity. They bring expertise in the study of these patients in parallel with animal studies. In particular their expertise in automated electrochemiluminescence and multiplex ELISA for the determination of cytokines and adipokines will be of importance for LipidomicNet.
